The move to the north London sees the Kenyan international reunite with Mauricio Pochettino. The pair worked together at Southampton in 2013/14.
The 24-year-old will add much needed strength to the Spurs squad that will see the club return to the UEFA Champions League after a five-year gap.
Wanyama has racked up 97 appearances in all competitions for the Saints, helping the club to achieve three top-eight finishes in the English Premier League.
